What's the right Semax dose: nasal spray or injection?
Most people running Semax start with 200 to 600 mcg a day through a nasal spray, split into one or two doses. Injection protocols use less per dose, usually 100 to 300 mcg subcutaneously, meaning under the skin, once or twice daily. Nasal spray is the route most people pick.

Why does nasal delivery reach your brain faster?
Your nose sits closer to your brain than your bloodstream does, structurally speaking. When you spray Semax into your nose, it crosses into brain tissue within about two minutes. It travels along the same nerve pathways your sense of smell uses, the olfactory and trigeminal nerves (Shevchenko 2006). Cerebrospinal fluid is the fluid that surrounds the brain and spinal cord. In animal studies, Semax reaches 60 to 70% of the matching blood level within about 30 minutes.
An injection has to clear your liver first. This process is called first-pass metabolism. It breaks down a portion of most drugs before they ever reach general circulation. Only a small fraction of an injected dose crosses into your brain afterward. That is why nasal spray produces stronger cognitive effects at a lower nominal dose. The number on the bottle matters less than how much actually reaches your brain.
Semax also breaks down fast in your blood. Plasma half-life, meaning the time it takes half the dose to break down, is about 2 to 3 minutes. But the effects last far longer, 12 to 24 hours or more. That's because Semax breaks down into an active byproduct called Pro-Gly-Pro. This byproduct stays in brain tissue and keeps signalling. Which route should you actually pick?
Pick nasal spray if cognitive performance, focus, or mood support is your goal. It is the route with the actual clinical evidence behind it, and it gets more Semax to your brain per microgram spent.
Pick injection only if nasal irritation stops you from tolerating the spray. You might also pick it if you already run an injection-based stack. This lets you add Semax without using a new delivery method. Just know that you are trading brain-targeted delivery for convenience.

How long should you run a Semax cycle?
Most community protocols run 2 to 4 weeks on, then take an equal break. Some run shorter 5 to 7 day courses for a specific push, like exam prep or a heavy work sprint. Russian clinical stroke protocols ran 10-day courses with a 20-day gap and did not taper.
Nobody has formally studied whether your brain builds tolerance to Semax with continuous use. The break is a precaution, not a documented requirement. A reasonable starting frame is two weeks on, two weeks off, adjusted to how you respond.
What to expect week by week on Semax
Week 1: acute focus and mental clarity, often within 15 to 30 minutes of a nasal dose. Some people notice a difference on day one.
Weeks 2 to 4 bring deeper effects on memory and mood. These effects build up as BDNF-driven changes add up. This time window shows the strongest signal in mood research. That includes a 2024 rodent study on stress resilience (Inozemtseva 2024).
